dc.description.abstractThe purpose of this project was to design and synthesise novel classes of mast cell stabilisers from benzocycloalkanones. This thesis is introduced with an overview of the increasing prevalence of allergic conditions throughout the world, and the fiindamental role mast cells and their associated mediators play in allergic reactions. The mechanism of the interaction between the antibody, immunoglobulin E (IgE), and its cell surface high affinity IgE-Fc receptors (FcεRI) is outlined. There then follows a review of numerous preformed and newly synthesized inflammatory mediators which are released by the activated mast cells following allergen subsequent exposure. This leads on to a discussion of the consequent pathological effects of these biological mediators in; allergic diseases, autoimmune disorders and inflammatory conditions. Chapter 1 concludes with an assessment of both currently available and investigational therapeutic options in the treatment of these mast cell-mediated diseases.
